The cup is in honour of late former Dimboola president and life member John Saxton and late Minyip-Murtoa life member and club stalwart Pat Walsh.
Wimmera Football League club Dimboola has the home-ground advantage in the second year of the cup competition.
President Alan Braithwaite said Dimboola's five-point win against Minyip-Murtoa in April would give the team confidence going into tomorrow's match.
"It will be an absolute ripper," he said.
"It has to be an advantage being on our own turf. We want to do John proud and bring it home."
Mr Braithwaite said Mr Saxton, who died of cancer in later 2008, was a great asset to the club.
He said the cup was a fantastic memorial to Mr Saxton and Mr Walsh, from Minyip Murtoa.
"The cup is in honour and memory of two great men in the footy clubs and their local community," Mr Braithwaite said.
Minyip-Murtoa won the inaugural cup match last year with a huge 71-point win.
Minyip-Murtoa Football Club president Brett Schmidt said Mr Saxton and Mr Walsh were legends of their respective clubs.
"It was a great honour to win it last year in its inaugural year," he said.
Mr Schmidt said Pat Walsh's wife Dianne would talk to Minyip- Murtoa players before they took to the field tomorrow.
"It's a great way of commemorating those guys," he said.
"Pat was a fantastic guy to have around the club and he is sadly missed."
Mr Schmidt said Minyip-Murtoa would go out fighting.
"Obviously it's a must-win game for us," he said.
"Dimboola just pipped us in the first round against them in the dying minutes so we would certainly love to turn the table."
Mrs Walsh said she would give players a pep talk before they took to the ground.
"I think the boys can do it again. They're a good team and they have a lot to play for," she said.
"The cup means a lot to them and it means a lot to the Minyip-Murtoa footy group."
Mrs Walsh said she had family travelling to Dimboola to watch the match with her tomorrow.
"I think I could speak on behalf of Jo Saxton as well in saying that we're extremely proud this cup has been put into action and it does keep our husband's memories alive," she said.
"They were so passionate about their football clubs. I'm very proud this is an ongoing thing."
