Ms Roll was last seen at a home in Drummond Street, Horsham and was reported missing on Wednesday.
Senior Sergeant Brendan Broadbent said Ms Roll's car was found in the Wimmera.
He was unable to specify where.
Sen Sgt Broadbent said police were searching the area.
"We are hopeful that she is still in the area,'' he said.
Horsham Sergeant Bill Alford said to disappear was out of character for Ms Roll.
Police are concerned for Ms Roll's welfare. She has a medical condition which requires regular medication.
She is described as being Caucasian, about 167 centimetres tall and of a medium build.
Police do not know what she was wearing when she disappeared.
They urge anyone who knows of Ms Roll's whereabouts to call Triple 0.
The Mail-Times recognised Ms Roll for her volunteering efforts in 2011.
She has been heavily involved with Holy Trinity Lutheran School, including organising the school's annual Christmas tree festival.
She has also volunteered with the Western Bulldogs Wimmera Supporters Group, Horsham United Football Club - where she was netball director for the 2010 season - and 2nd Horsham Scouts.
