The State Coroner, Victoria Police Disaster Victim Identification specialists and accident investigators are working to identify the two drivers, who were killed in a horror crash near Stawell at 11.30pm.
Police media officer Natalie Webster said police would contact the families involved as soon as investigations were complete.
She said many people had called police asking who was involved in the crash.
Ms Webster urged people concerned about family members to call their employer first.
She said only trucking companies should call Ararat Police Station about the crash, on 5355 1500.
Police will prepare a report for the coroner.
Another man, in his 50s, is in a serious but stable condition at The Alfred hospital in Melbourne.
The Western Highway will remain closed for most of the day as the investigation continues.
