The Roos, under coach Stuart Farr, have strengthened their list for 2014 and will be up against a developing Saints side under the tutelage of new coach Shayne Breuer.
Both sides will be looking to start their season on the right note and both coaches have said last season's grand final result is now in the past.
"It starts again it doesn't matter who you play," Farr said.
"Hopefully we'll get to play our style of footy and on our home ground we can make it hard for our opposition."
Farr said new recruits Shaun McDonald, Tyler Blake, Cam Ellis, Jeremy Pekin, Simon Clugston and up-and-coming junior Jayden Kuhne were set to play.
However, he said the side would not be at full strength in round one.
"We've got a few out but it's a good opportunity for a few guys who have had big pre-seasons," Farr said.
The boys from Coughlin Park have added to their squad over the past few weeks with the additions of players including Daniel Rees, from Central Highlands Football League, and Tom Pain, from Bendigo Football League.
Breuer said while he was wary of the Roos, the Saints would be focusing on what they could control.
"We'll be trying to get our structures right," he said.
"The majority of our focus will be on ourselves and playing the way we want to play.
"We've got a few new guys and a few young guys coming through, so hopefully they gel together."
