The classic is a walk or run and is a major fundraiser for the National Breast Cancer Foundation.
Ararat, Donald, Horsham, Rupanyup and Stawell will all host events this year.
Horsham organiser Hanne Mills said it was the fifth year the city had run the event.
She hoped more than 500 people will support the event, which will start and finish at Horsham Soundshell.
Mrs Mills said people could register for the May 11 event online at www.mothersdayclassic.com.au or on the day.
She said people who registered online would go in a draw to win a morganite and diamond rose gold dress ring worth $1499. Mackays Leading Edge Jewellers in Horsham donated the ring.
The event costs $20 for adults, $12 for students and health card holders, $10 for children, $50 for a family of two adults and up to four children, and $30 for a family of one adult and up to four children.
Registration for the Horsham, Rupanyup and Stawell events starts at 8am, while the Donald registration is from 8.15am and Ararat’s from 9am.
The events start at various times.
Mrs Mills said Horsham’s event would include warm-ups, a guest speaker, remembrance and balloon releases before the walk.
She said people could complete an eight-kilometre run or a four-kilometre walk.
Mrs Mills said people could visit the Mother’s Day Classic website for more event details.
