Leeanne Nuske is an enrolled nurse and supervisor at Eventide's Warne House and colleague Andrea Monaghan is a cook there.
The pair will walk 466 kilometres along the St James pilgrimage from Leon to Cape Finisterre with the aim to raise $4660 or $10 a kilometre.
Ms Nuske said the pair would start on Saturday next week.
She said they aimed to complete the trek in 19 days.
The walk finishes at the Cathedral of Santiago de Compostela, where the remains of apostle St James the Great are believed to rest.
Ms Nuske said thousands of pilgrims completed the trek to the cathedral each year.
She said as long-time employees at Eventide Homes, she and Ms Monaghan could not think of a more special way to raise money for it.
"The journey will be very special for us,'' she said.
Ms Monaghan said she looked forward to meeting other pilgrims along the way.
She said the pair would mainly stay at pilgrim hostels known as albergues.
Eventide Homes chief executive Sue Blakey encouraged people to support the women's efforts.
She said money raised would go towards iPads for residents to use.
Ms Blakey said Eventide would have a map so residents could track the women's journey.
"I think it's going to be really interesting for Eventide residents and the broader community to track what will be an impressive effort by Leeanne and Andrea,'' she said.
She said people could donate towards the cause at www.mycause.com.au/page/76272 or at Eventides Homes reception.
