Leading Senior Constable Matt Haughton said a Naracoorte couple was towing a trailer when its vehicle left the road and crashed into a tree about 1pm on Monday.
The male driver and female passenger were taken to Wimmera Base Hospital by ambulance.
Leading Sen Const Haughton said they both had moderate injuries.
He said they were travelling west about five kilometres outside of Horsham when the crash occurred.
"The vehicle had extensive damage," he said.
Leading Sen Const Haughton said the cause of the accident was being investigated.
"Fatigue is being investigated as the cause," he said.
"Driving conditions were good at the time."
Ambulance, police, Country Fire Authority and State Emergency Service crews attended the scene.
