Team Jaam Jaam will compete in the Melbourne Ride to Conquer Cancer on October 25 and 26.
Money raised goes to the Peter MacCallum Centre, Australia's only public hospital dedicated to cancer.
Team Jaam Jaam captain Cass Hebbard said the team had two riders last year.
This year there are 16 riders, with 13 of those from Halls Gap.
Miss Hebbard, who lost her Gran to cancer, said the team hoped to raise $49,500.
They have raised more than $10,000 so far.
"This will be my second year riding in honour of those in my life affected my cancer," Miss Hebbard said.
"After last year's ride I decided I would love to raise more money for Peter Mac to support them on their epic quest to conquer cancer.
"My thought was simple: 'Many hands make light work' - a phrase my Gran would often quote.
"So, I set about spreading the word and creating a larger team."
Participants will ride from Melbourne to the Mornington Peninsula over the two days.
Miss Hebbard said the word Jaam was Nepalese for 'let's go'.
She said Team Jaam Jaam was one of the largest joining in the ride.
"We are a team of highly passionate, enthusiastic and determined people with a range of incredible stories and backgrounds," she said.
"We all love to laugh and very rarely do the smiles disappear from our faces."
Miss Hebbard encouraged people to donate to the team to help them reach their goal.
She said people could visit the team Facebook page 'TeamJaamJaam' or email casshebbard@hotmail.com for information on how to donate.
Miss Hebbard said the team was fortunate to have five sponsors so far, including Halls Gap Tourism, Stawell and Grampians Towing, Coolas Ice Creamery, Comfort Inn Country Plaza and Halls Gap Fair Price Chemist.
The Ride to Conquer Cancer has raised more than $10 million toward projects and services for Peter Mac in the past two years.
