This follows the arrest of 38-year-old Trudy Giles, also known as Trudy Gilson, who appeared before Port Macquarie Local Court on Saturday charged with the alleged stabbing murder of a 43-year-old man near King Creek.
Police and paramedics were called to a car accident along the Oxley Highway east of Wauchope at 7am on Friday morning where they found the injured man inside his car.
Police allege the accused met the man near King Creek at about 4.30am before she stabbed him once in the chest. It is alleged the man then drove off.
The man was treated by paramedics on the roadside before dying en route to Port Macquarie Base Hospital.
Giles/Gilson appeared before the court on Saturday and has been refused bail.
Police from the Mid North Coast Local Area Command continued investigations into the matter on Saturday and executed a search warrant at an address in Pappinbarra.
During the search police seized a knife, clothing and approximately 50 cannabis plants.
At about 10am Saturday, police arrested a 43-year-old man in Murray Street, Port Macquarie.
The Pappinbarra man was taken to Port Macquarie Police Station where he was charged with accessory after the fact to murder and cultivate cannabis.
He was refused bail to appear in Port Macquarie Local Court on Sunday.
SATURDAY 10am: A 38-year-old woman has appeared in Port Macquarie Local Court charged with murder.
Trudy Giles also known as Trudy Gilson fronted court on Saturday morning.
Police said Giles/Gilson was charged with the alleged murder of a man who was found in a car near Wauchope on Friday.
Police allege the accused met with a 43-year-old man near King Creek on Friday morning. The two knew each other, police allege.
Police will allege the woman stabbed the man in the chest, before he got into his vehicle and drove off.
Just after 7am,police received reports of a man injured in a car on the side of the Oxley Highway at Wauchope.
Paramedics treated him by the roadside but he died on the way to hospital.
After making inquiries, police arrested a 38-year-old woman at a Park Street address in Port Macquarie.
Police executed a search warrant at this address, where they seized cannabis plants and other items. A second search warrant was also executed at a Pappinbarra address.
The woman was taken to Port Macquarie Police Station where she was charged with murder.
She was refused bail.

A MAN has died after an incident on the Oxley Highway east of Wauchope on Friday that left his car in flames on the side of the road.
Senior police officers have been at the scene throughout the day investigating the incident and have declared the man's death as suspicious.
A spokesperson from the Police Media Unit in Sydney confirmed the accident occurred just after 7am on Friday on the stretch of Oxley Highway between King Creek Road and the entrance to Wauchope.
Officers from the Mid North Coast Local Area Command were called to the scene after reports there was a man injured inside the car.
On arrival, police and paramedics found a 43-year-old man in a Ford Falcon sedan which had been engulfed by flames.
Paramedics treated the man at the scene but he died on the way to Port Macquarie Base Hospital.
Residents at the northern end of Narran River Road reported hearing the sound of screeching before seeing black smoke billowing from the accident scene.
Senior officers continue to investigate the circumstances leading up to the man's death.
