Police believe the man had been crossing the road about 10.30pm on Sunday night when he was hit by an east-bound Holden Commodore.
Witnesses attempted to perform CPR but the man, aged in his 30s, went into cardiac arrest, an Ambulance Victoria spokesman said.
He died at the scene.
Police said the man had been a passenger in a Ford Falcon that had pulled over near the Tullamarine Freeway interchange.
The drivers of both vehicles are assisting police with inquiries.
