The club will host this year's 51st annual show, also known as Gemboree, at the Wimmera Events Centre at Longerenong from Friday.
Club president Brian Mann said hundreds of people had already registered to stay on site.
He said many more had booked accommodation throughout the region.
"Now in its 51st year, this show has been hosted in Victoria only eight times," he said.
"We hosted a great show here in Horsham in 2009 and I am quietly confident this one will be even better.
"We have a great little club here in Horsham with members who are not afraid to volunteer when there is work to be done."
Mr Mann said Horsham Rural City Council, Wimmera Events Centre and Wimmera media and sponsors supported the event.
"We have been able to assist the organisers, the Victorian Gem Clubs Association, in putting together an event that will hopefully be the envy of the other states," he said.
"There is still work to be done, but we are on the home straight now."
Mr Mann said the Moore Exhibition Centre would feature a comprehensive collection of gemstones, jewellery, minerals, fossils and association products.
More than 1000 national competition entries of cut and polished gemstones, jewellery and allied crafts will also be on display.
"It will be both exciting and educational," Mr Mann said.
"Whether people come to buy or just to browse, this is a special opportunity they won't want to miss."
Victorian Gem Clubs Association secretary Bill Gordon said the Horsham club was a fine example of what dedicated volunteers could achieve in a supportive community environment.
"Their hard work has certainly set the stage for a brilliant Gemboree," he said.
The Gemboree will kick off on Friday from noon to 5pm and continue on Saturday and Sunday from 9.30am to 5pm and Monday from 9.30am to 2pm.
Mr Mann said day tickets were $20 and single entry and concession prices were available.
