The store is located in the city's central business district at 81 Firebrace Street.
Major Carolyn Wright said adding a second store to the city made their service more accessible to Horsham residents.
She said not everyone could get to the store on Lynott Street.
"It's to serve people who don't go over the bridge," she said.
Major Wright said the new store was a great find.
"It is a big location - it is bright and modern," she said.
Salvation Army thrift shops sell pre-loved clothing, bric-a-brac and furniture to support the organisation's community programs.
The organisation aims for sustainability conscious stores.
Major Wright said donated items would now be shared between the two stores, with items from the Lynott Street site moved to the Firebrace Street site for the opening.
She said the stores relied on donations and volunteers.
"We'll be looking for as many volunteers as we can," she said.
She urged anyone interested to volunteer.
Major Wright said people could volunteer as much time as they wished, from one shift per week or longer stints from Monday to Saturday.
She said volunteers did not need any prior experience.
"We can train them on the spot," she said.
Major Wright said the organisation hoped to offer more than just shopping opportunities in the new store.
"We're hoping to have some different things," she said.
She said the service planned to set up a corner of the store for people to sit and browse through books in a relaxed setting.
Major Wright said the organisation hoped to organise a childrens reading hour.
"Children can hear a story while their mums are shopping," she said.
She said the official opening would be Friday at 10am with members from Salvation Army attending.
