The comedian, whose Australian tour launches in Sydney on Saturday, took offence to claims Armytage had labelled her as "unwell" following a recent run-in on the Seven morning show.
"She's been doing all these interviews... I kinda put her on the map," Griffin told radio hosts Kyle & Jackie O.
"I'm happy to challenge her to a cage match. Two dogs enter, one dog leaves," she said, adding she fancies her chances.
"I'm little, but I'm scrappy... I'm a float like a butterfly, sting like a bee," she joked.
Griffin and Armytage clashed during the comedian's appearance on Sunrise in August, over Griffin's controversial stunt where she posed with a "severed head" of Donald Trump.
The Seven host drew Griffin's ire when she implied the public fallout to the stunt - which saw Griffin fired from a regular gig on CNN, and effectively blacklisted in Hollywood - was justified.
"No, you're full of crap!" Griffin told Armytage at the time.
"Stop acting like my little picture is more important than talking about the actual atrocities that the president of the United States is committing," she said.
Reps for Sunrise and Armytage did not immediately respond to request for comment.
Griffin also doubled down on her criticism of Trump, telling Kyle & Jackie O, "I'm just gonna call it like it is - there's a Nazi in the White House, and we're all scared.
"Once he started doing all the heinous acts that he's doing, that's when I went, 'Okay, screw the people that got mad 'cause I took a picture with a children's Halloween mask and put ketchup on it...'
"I had to be in a two-month federal investigation, be on the no-fly list... I had to do the perp walk and talk to the feds... I'm gonna let it all out on stage," she said.
Griffin's Australian tour, which includes a show at the Sydney Opera House and a sold out gig in Melbourne, wraps up in Perth on October 31.
