McDonald was initially drafted as a ruckman to Jack Riewoldt's Rampage side for the tournament at Marvel Stadium, but will be rested following an injection in his knee to relieve soreness.
The Demons confirmed on Wednesday that McDonald had been administered a cortisone injection to relieve patella tendinitis, but is not expected to miss any other games.
McDonald joins a growing list of AFL stars who were initially drafted to take part in the AFLX showcase but have since pulled out due to injury.
The much-maligned concept has come under further scrutiny with nine drafted players no longer going to participate in the tournament.
McDonald was replaced in the side by Melbourne teammate Bailey Fritsch.
