Craig Bennett, Geoffrey Dumesny, Craig Garland, Frank Giampaolo and David McCluskey are part-owners of Nature Strip, who will compete for the $14 million prizemoney in the day's headline race.
Mr Garland said it was an intense build-up.
"For five guys from Horsham to have a spot in the Everest, it's pretty incredible - it's just about as good as having a runner in the Melbourne Cup really," Mr Garland said.
"We're pretty nervous but excited ... there's a fair build up at the moment."
Mr Garland said following Nature Strip's journey since they acquired a stake in the horse had been an incredible experience.
The horse qualified for the Everest last year, but was a late exclusion after a poor run before the event.
"It ran a shocker in the lead-up last year and (previous trainer) Darren Weir just sent it straight to the paddock," Mr Garland said.
"That was a bit of a let down ... but now to be back it's just all excitement.
"Two years ago in the week leading up to the first Everest, we said, 'wouldn't it be nice to be in the Everest one day?' And here we are.
"It's been a magnificent journey. Let's just hope that it can put in a great effort."
Nature Strip has won two Group 1 races this year, including the Galaxy in Autumn and the Moir last month.
The biggest question marks are whether it can run out the full 1200 metres. The horse was at $26 with most betting agencies on Friday afternoon.
"It's going to be a fair task," Mr Garland said.
"Hopefully we can just be coming around the corner sitting alongside Redzel and Sunlight with 50 metres left. Wouldn't that be incredible?"
