The historic building, located west of Goroke, is believed to be built before 1864, according to Heritage Council Victoria.
The two-storey homestead, and accompanying meathouse and stables, was built for David Affleck.
The National Trust-listed building is believed to have been demolished within the last fortnight.
The owner of the property has been contacted for comment.
An anonymous resident told the Mail-Times they was disappointed to hear about the historic building's demolition.
The resident said the building was "falling down to a certain degree" before its demolition.
"I was speechless when I was told," they said.
"It was a pretty spectacular building - you see pictures from inside the homestead of great big doors, the magnificent staircase, all that sort of stuff.
"It was just a significant, wonderful, historic homestead.
"It is a shame."
Heritage Council Victoria, the National Trust of Australia, and the West Wimmera Shire have also been contacted for comment.
