Dimboola Sergeant Steve Gray said four Country Fire Authority units were called to Lower Roy Street at 3pm.
"We ended up having crime scene guard there overnight because we couldn't determine whether it was suspicious before it got dark," he said.
"There was one person at home at the time, in the back shed, and the house became fully engulfed. The man suffered smoke inhalation, but I don't believe he was taken to hospital.
"The police member up there arranged accommodation for the family of four that lived there through Uniting Wimmera."
Sergeant Gray said a member from Victoria Police's Melbourne-based Arson and Explosives Squad was set to investigate the fire on Friday morning.
