Northern Territory superstar Shannon Rioli and 2019 North Central league Feeny medallist Mitch Gleeson headline the list of players re-committing to the Panthers.
Former North Ballarat Rooster Jarryd Graham has also re-signed.
The trio of recruits signed with the club prior to the cancelled 2020 season, but are yet to pull on the blue and red in the Horsham District football league.
Rupanyup coach Jack Musgrove said the club was pleased to have retained the services of the talented group.
"We've worked pretty hard on getting them back on board the last six months and we're stoked that they've all signed back on," he said.
"It was very disappointing we didn't get a chance to show them off last year, but they're all really good blokes and they were all keen to come again for another year, which is great."
Fellow Darwin recruit Jimmy McMurtrie, and former Panthers Brett Niewand, Chris Schaper and Cam Ruwoldt are also on board.
Musgrove said the re-signing crop provided some "top end talent".
"The blokes that we've got are all pretty handy," he said.
"Shannon Rioli has won a lot of flags up in Darwin, and is a very high profile player, so we're really looking forward to having him at the club," he said.
"Mitch Gleeson is a massive signing as well - he's played a bit in the ruck before, but he might just slot into the midfield as a big-bodied mid which will provide a lot of help.
"Jarryd Graham has also tasted success at a really high level, he's a really hard nut midfielder so we're rapt to get him as well."

Musgrove said there could also be more signings to come for the club.
He said with an improved playing list, Rupanyup had a clear goal in 2021 to play finals for the first time since 2012.
"At the end of the day, we want to have a bit of success as a footy club," Musgrove said.
"Having these high-end talent guys gives us some leadership.
"With these signings, we've got the aim of playing finals. We haven't done that in a long time, so just getting there and playing finals, that is our biggest thing."
