Deputy premier James Merlino also announced the Ballarat lockdown will end at midnight on Wednesday as planned.
"The public health team feels the outbreak is contained," he said. "Thanks to residents who came out in droves to be tested and followed lockdown rules.
"Ballarat's settings will align with areas in regional Victoria that are not in lockdown."
Mr Merlino also said the chief health officer had approved areas of Point Lonsdale would be able to follow the same rules as the Borough of Queenscliff from 11.59pm on Wednesday.
Deputy chief health Deb Friedman said along with the Macedon Ranges case, new regional Victorian cases were found in Wangaratta (1), East Gippsland (1), Geelong (2), Mitchell Shire (4) and Mooroobool Shire.
Dr Friedman said the source of the Macedon Ranges case was unclear and is being investigated.
"We are pleased to end lockdown in Ballarat but there is a positive case that resides in Melbourne but worked at the GovHub in Ballarat," she said. "That person was an acquisition source for a case in Ballarat that also worked at the GovHub.
"The GovHub is a tier 1 exposure site for the period between September 13 to 17. All staff who were present are in quarantine and are getting tested. The site is closed but no impacts to services are expected as staff are working from home."
The department this morning published new statistics showing 60,829 people were tested for COVID-19 on Tuesday and another 43,056 were vaccinated.
Active cases in Victoria now total 6223.
Bendigo Health is also reminding people they only need to be tested if they were at exposure sites on days and times listed by the Department of Health.
"You do not need to get tested if you were not personally at the exposure site," the group said in a Facebook post.
