South Australia police said two trucks collided on the Dukes Highway, about five kilometres west of the Victorian border.
Both drivers were injured.
Police and emergency service crews have closed the highway, with vehicles being diverted at Keith onto the Riddoch Highway to Naracoorte, then to Horsham via the Wimmera Highway.
Diversions are expected to be in place until about 2pm.
The crash is the latest in a series of accidents this week.
A truck clipped a Horsham power line on Tuesday, causing power cuts to more than 3000 customers.
Late yesterday a mini bus full of students rolled over in the Grampians. An air-ambulance flew one school-girl to Stawell District Hospital.
