The Panthers won their third flag and their first for a decade with a 32-point win over Murtoa in the Wimmera Football League 40 years ago.
Since joining the Horsham District Football League in 1981, the team has won a further seven premierships.
The club's function will follow Rupanyup's home clash with Laharum this weekend.
Reunion organiser and Rupanyup premiership player Kevin Thewlis said 17 members of the side were expected to attend.
He said the club was hoping for a big attendance at the game and the function.
"We're trying to get back as many supporters and former players of that era as we can to renew old acquaintances," he said.
"It's a pretty big event for the club and it would be nice to have as many there as possible." The team featured several prominent Wimmera football players such as former South Melbourne VFL full forward John Sudholz and an 18-year-old Greg Bell, who went on to play with Essendon.
The side was coached jointly by Steve Arnott and Max Matheson.
"There was a lot of quality in the side that year," Thewlis said.
It was hard to re-assemble most of the team, with only two players still living in the area, he said.
