Horsham Acting Sergeant Cass Myers said the Horsham woman was driving east along O'Callaghan Parade when she failed to give way at the roundabout at the Stawell Road intersection.
Police and fire crews were on the scene near the BP Wimmera Bridge Service Station about 11am.
"Her sedan collided with the front of a B-double truck that was travelling north," Acting Sgt Myers said.
"There was no alcohol involved - we believe it was driver error."
Acting Sgt Myers said the woman would receive a penalty notice for failing to give way.
State Emergency Service and paramedics also attended the crash.
Emergency service Horsham unit controller Jason Woodhart said everyone had escaped the vehicles by the time the service arrived.
