Now in its eighth year, the national initiative celebrates making the most of the great outdoors and encourages non-cyclists to try riding.
Holy Trinity Lutheran School principal Jeff Gork said about 140 students from the Horsham school took part.
"It's the most participants we've had in the four to five years we've been doing it," he said.
"They loved it. We had quite a few students in prep and grades one and two who were riding to school for the first time.
"It was such a beautiful day they were beaming."
Mr Gork said Ride2School Day helped promote cycling as a fun activity.
"It's part of our focus on the overall well-being of students," he said.
"We're not just concerned with academics, but also with the students' physical and emotional well-being."
Mr Gork said the school would continue to participate in Ride2School Day. He hopes the initiative will encourage the children to continue cycling.
"The parents were very happy to see their students riding to school," he said.
Mr Gork said three groups of students, each led by staff, converged on the school from different directions.
"To see all the students riding down Trinity Drive as a big group was a great sight to see," he said.
