A Victoria Police spokeswoman told the Mail-Times a truck carrying livestock collided with a white Mazda SUV on Stawell-Warracknabeal Road about 11.45am.
"A passenger of the SUV, a 34-year-old South Australian woman was flown to hospital with serious but non-life-threatening injuries," she said.
"Two other occupants of the vehicle, 32-year-old South Australian man and a 31-year-old South Australian woman, were transported to hospital with minor injuries.
"The truck driver, an 35-year-old Hillside man, was transported to hospital with minor injuries. He is currently assisting police with their inquiries."
A number of livestock were euthanised at the scene.
EARLIER: One person is recovering from serious injuries in Royal Melbourne Hospital, after a sheep transport truck and a car collided near Minyip on Sunday.
Ambulance Victoria has confirmed it was called to the accident, which was reportedly on a stretch of Stawell-Warracknabeal road near the town around 11.45am.
"Four patients were transported to Wimmera Base Hospital in road ambulances with upper body injuries and all four patients were in a stable condition," a spokeswoman said.
"One patient was taken by air ambulance helicopter to the Royal Melbourne Hospital in a serious but stable condition and appears to have upper body injuries and abrasions."
Witness reports on social media suggested the incident left the sheep truck on its side and some livestock injured.
Warracknabeal's SES Unit attended along with police. Unit controller John Bish said he understood the two vehicles were travelling in opposite directions, and volunteers extricated four people from the car, believed to be an SUV.
He could not confirm whether the truck driver needed to be freed.
