Horsham Highway Patrol stopped the man, 49, and the woman, 46, on Wednesday about 1pm at Miram, between Kaniva and Nhill.
Sergeant Dale McIvor said police found "a large amount" of cannabis and cash in their car.
The pair will face court in Horsham on Thursday.
Sergeant McIvor said it was a "terrific find".
"Members conducted a search of the vehicle and found a large amount of cannabis and a large amount of cash," he said.
"The male driver and the female passenger were arrested and brought back to the Horsham Police Station, as well as the vehicle.
"It was a really awesome pinch ... a really good find by our highway patrol members."
