More than 50 bands and musicians will perform at four venues across Horsham from February 7 to 9 - the Horsham Town Hall, Horsham Town Hall's Heritage Hall, Maydale Pavilion at the Horsham Showgrounds and Horsham Soundshell at Sawyer Park.
Bands performing at the festival include Shades of Troopers Creek, RIFT, Second Thought/Circus Animals, All the Kings Men and Acoustics Stomp.
The event comes more than a decade after 50 Years of Wimmera Rock, which saw 45 bands play around Horsham as part of the 2009 Kannamaroo Festival.
Event committee member Dave McMaster said many bands had reunited for this year's special anniversary event.
"There is so much choice - everything from folk to heavy metal, we've got it all covered," he said.
"The 50 Years of Wimmera Rock was a huge success 10 years ago. It was a big event for the whole community of Horsham and a big reunion of old bandmates getting together.
"We hope this year's will be bigger and better than that one."
Mr McMaster's own band RIFT will perform from 10pm on Saturday in Horsham Town Hall's Heritage Hall.
Horsham Town Hall marketing officer Charee Smith said performances would span six decades and many different of genres of music.
"Many musicians are returning to Horsham since the redeveloped Horsham Town Hall has opened, all with fond memories of performing on the Heritage Hall and the many pubs and clubs around the region," she said.
"The city's vibrancy will have an incredible soundtrack as many bands have been booked by hotels such as Bull 'n' Mouth, Exchange Hotel, the RSL and Westside Cabaret."

Horsham Riverside Caravan Park co-manager Sue Jones said sites and cabins at the park were almost sold out for this weekend.
"We've had a lot of interest from people who are coming for the festival. We're not completely sold out yet, but I think a lot of people will be leaving their bookings to the last minute," she said.
"We're in a really good location for the festival being right next to the soundshell; I think it will be a great event for the town and hope it's a great success."
