South Australia's chief health officer Nicola Spurrier announced the border ban at an afternoon press conference.
Victorian cross border community members will not be affected by the ban, with anyone living within 70km of the border exempt from the closure.
This comes after two teenagers breached South Australia's border checkpoints and snuck into the state.
The two young girls were detained and tested negative for COVID-19.
EARLIER: South Australian premier Steven Marshall has announced anyone from Greater Melbourne will not be permitted to travel into South Australia from 6pm today.
At an afternoon press conference South Australian chief health officer Nicola Spurrier said she was unsure of how long the restriction would be in place, due to the sudden nature of the positive COVID-19 cases.
Cross border residents from regional Victorians were not included in the Premier's travel ban; however, this may change after Ballarat Health Services announces a potential COVID-19 cases was "under investigation".
The announcement comes after South Australian authorities confirmed a Melbourne man contracted COVID-19 in hotel quarantine in Adelaide, likely through aerosol transmission.
The person is being monitored at home, but where the person returned from has not been released.
Meanwhile, in a move that is a boon to regional Victorians, the Therapeutic Goods Administration changed its storage conditions for the Pfizer vaccine, allowing the vaccine to be stored longer at normal refrigeration temperatures.

MAY 26: The South Australian government has introduced new restrictions for anyone entering the state from Victoria.
With four new cases confirmed, SA will stop travellers who visited a number of locations in Melbourne's north from travelling to SA.
Under the new restrictions, which came into effect at 8.45pm May 24, anyone who has been at a public exposure site will be prohibited entry into South Australia unless they are a domestic violence arrival.
Anyone already in South Australia who visited the City of Whittlesea on or after May 6, 2021, will have to self-quarantine until a negative COVID-19 test.

Travellers will still need to complete a Cross Border Travel registration form at least seven days before arrival in the state.
On Monday, South Australia's Chief Public Health Officer Nicola Spurrier said she had received a briefing from Victorian officials.
"I'm just working through the implications of that. Victoria's contact tracing team are obviously doing an excellent job," she said.
Premier Steven Marshall said while SA was concerned by the situation in Victoria, local officials had a good working relationship with their interstate counterparts.
"Any new case in the community is a concern to us. We've got to do everything we can to keep ourselves protected," the premier said.
