Senior Constable Adrian Hoggett said an office at the depot was broken into sometime between 7pm on Friday and 10am on Saturday.
"A significant quantity of cash was taken from the office's social club money," he said.
Senior Constable Hoggett said it was disappointing the depot was targeted.
The burglary comes after a spate of vandalism in the town.
Senior Constable Hoggett said infrastructure at the town's Band Park was damaged earlier this month.
"At the children's playground, the gates were torn off the hinges," he said.
"They have now been replaced twice, once on Friday, and again on Saturday."
"It's quite disappointing and we seem to have had a spate of incidents in the town over the past week or so," Senior Constable Hoggett said.
Police have urged for anyone with information on either the burglary or the vandalism to come forward.
Senior Constable Hoggett said people should call the Kaniva Police Station on 5392 2244.
